IN a recent American television broadcast that left many gaping with amazement, famed American celebrity doctor Mehmet Oz, better known as Dr Oz, actually sang the praises of palm oil. Specifically, the doctor was extolling the virtues of red palm oil (RPO), which in industry parlance would be the crude oil extracted from carefully selected fresh fruit bunches of the oil palm tree ( Elaeis guineensis ). Friday 7th. Dec 2007 Today is the first day I stain my slides. What I did today 5 slides all together - diltion: 1/50, 1/250, 1/500, one slide with Hx alone, one slide with secondary alone Water bath - It took 15 minutes to increase 10 0 C. Dewaxing - Hot plate 60 0 C - 15 minutes Rehydration - Xylene 2x, Alcohol 100 % 2x, 90 % , 80%, 70%. 50 % not used. Antigen retrieval solution in jar - heated earlier Slides put into jar - temperature drop a bit when the cover was lifted up. Tempertaure pick up above 95 0 C in about 10 minutes. Slides in ARS for 30 minutes Cooling down - 30 minutes TBS - Shaker 3 min x 2 H2O2 - 10 minutes TBS shaker 3 min x2 Primary AB - 1 hour TBS Shaker 3 min x2 Secondary antibody - 30 minutes TBS shaker 3 min x2 Chromogen DAB 10 minutes TBS Shaker 3 min x2 wash with DH2O Hematoxylin 30 s..drip only at sink wash with running water Dehydrate - 70-80-90-100-100-xylene-xylene wipe dry DPX dry Monday baru boleh angkat

Plan untuk esok: Jumaat 7th. Dec 2007 On water bath. Letak antigen retrieval solution dalam bekas kaca. Letak dalam water bath bertutup On hot plate. Letak 4 slide ats hot plate at 60 o C. 30 minit Dilute kan DAB. Simpan dalam fridge De wax - xylene (2x), etanol 100-100-95-80-70-50. All 3 minit Letak dalam retrieval solution - 30 minit take out from water bath -cool at room temperature 20-30 minit wash titik H2O2 wash - belly dancer titik primary antibody atas 3 slide at 1/50, 1/250, 1/500 - satu slide tak payah primary sebagai kontrol. simpan balance primary dalam botol lain wash - belly dancer secondary wash - belly dancer DAB - chromogen wash -belly dancer hematoxylin DPX cover dgn slide kaca view

6th. Dec 2007 Today - dah dapat settlekan water bath. Can maintain tempertaure around 95-99 o C. Dah buat TBS baru - stock and diluted solution. Supervisor suruh buat homocysteine week 10 first once dapat kit. Need to publish 3 papers before grad....phew

5th December 2007 Immunohistochemistry: 1. primary antibody diluted: 1/50 ( add 10 ul antibody + 490 ul antibody diluent). Then 100ul taken and added to 400 ul antibody diluent= 1/500 dilution. Both kept in fridge together with antibody diluent and primary antibody 2. DAB and substrate not yet diluted. Will prepare fresh 3. For dewaxing: Xylene, alcohol 100%-50 % all prepared in bottles and in place. 4. Hematoxylin - set 5. DPX - set 6. Antigen retrieval solution - in fridge can reuse 10 x 7. Belly dancer: set 8. Hot plate : set 9. Gauze: set 10. Water bath: Gila betul water bath kat sini. Ada 3 water bath satu pun tak leh guna. Dari pukul 11.10 aku try increase temperature tak naik-naik sampai petang. Bila dah naik every 30 second kena click manually. Bila dah bukak , nak bubuh specimen temperature drop to 80's.....apa da. camne nak dapat temperature yg stabil nih. Last last aku yang menggelegak. Tak pe dah inform kak miah and kiv tumpang water bath biochem...tumpang lagi. So sang...
